Abstract

Udar mózgu jest jedną z głównych przyczyn śmiertelności w krajach wysoce rozwiniętych. Kluczem do opracowania skutecznej metody leczenia udaru jest lepsze zrozumienie patofizjologii choroby. Opracowanie tego fundamentu wymaga zastosowania specjalistycznych modeli zwierzęcych, które naśladują proces fizykochemiczny danej choroby. Jedną z ciekawych opcji medycznych, które mogą posłużyć leczeniu udaru jest teranostyka. Teranostyk to substancja łącząca w sobie modalności diagnostyki i terapii. Dzięki zastosowaniu teranostyków oraz wybranej metody obrazowania, możliwe jest dostarczenie środków terapeutycznych do konkretnego obszaru anatomicznego pod kontrolą techniki obrazowej. Celem niniejszej pracy było przeprowadzenie badań własności relaksacyjnych nowych teranostycznych nanonośników w warunkach in vitro przy użyciu obrazowania rezonansem magnetycznym MRI. Następnym celem była ocena możliwości efektywnego dostarczania środków neuroprotekcyjnych do mózgu, poprzez charakteryzację udaru mózgu, wykorzystując model zwierzęcy udaru niedokrwiennego MCAO (ang. Middle Cerebral Artery Occlusion). Kluczowymi aspektami w wykonanych badaniach były pomiary wartości czasów relaksacji T1 i T2, pomiary współczynnika dyfuzji w tkance mózgowej, badania przepuszczalności bariery krew-mózg oraz ocena limitu detekcji środka kontrastowego w tkance niedokrwiennej, co jest kluczowe w przypadku dostarczania środków neuroprotekcyjnych do mózgu pod kontrolą MRI. Przeprowadzone badania pozwoliły na ocenę potencjału teranostycznego szeregu różnych nowych nanocząstek. Ponadto badania pozwoliły na zoptymalizowanie indukcji modelu udaru MCAO. Dodatkowo pozwoliły one scharakteryzować tkankę niedokrwienną wykorzystując obrazowanie MRI. Badania pozwoliły na ocenę ewolucji czasowej udaru oraz ocenę przepuszczalności bariery krew-mózg. Niniejsza praca pozwoliła na zoptymalizowanie protokołów pomiarowych na cele badań biodystrybucji oraz wdrożenie zaawansowanych i autorskich programów do analizy danych obrazowych MRI. Uzyskane wyniki badań będą fundamentem w przyszłej ocenie efektów terapeutycznych różnych teranostyków.
Ischemic stroke is one of the biggest causes of mortality in highly developed countries. A better understanding of the pathophysiology of the stroke is key to developing an effective treatment strategy. Establishing this foundation requires the usage of specialized animal models, which mimic the physicochemical processes of the disease. One of the promising medical approaches for stroke treatment is theranostics. This concept combines diagnostic and therapeutic modalities into a single nanoplatform. By using theranostics along with selected imaging techniques, it is possible to deliver therapeutic compounds directly to specific anatomical regions under the guidance of medical imaging. The aim of this study was to investigate the relaxation properties of novel theranostic nanocarriers in vitro using magnetic resonance imaging MRI. Another objective was to evaluate the potential for efficient delivery of neuroprotective drugs to the brain by characterizing ischemic stroke using the MCAO (Middle Cerebral Artery Occlusion) animal model. Key aspects of that research included measurements of T1 and T2 relaxation times, diffusion coefficients in brain tissue, blood–brain barrier permeability assessments and the determination of the detection limit of the contrast agent in ischemic tissue, which are critical for MRI-guided neuroprotective delivery. The conducted studies enabled the assessment of the theranostic potential of a range of novel nanoparticles. Furthermore, these investigations facilitated the optimization of the MCAO stroke model induction and allowed for detailed characterization of ischemic tissue using MRI. The research also provided insights into the temporal evolution of stroke and the evaluation of blood-brain barrier permeability. This work has enabled the optimization of imaging protocols for biodistribution studies and the implementation of advanced, custom-built softwares for MRI image data analysis. The obtained results will serve as a foundation for the future evaluation of the therapeutic effects of various theranostic agents.
