Abstract
2% żel agarozowy przygotowano z 4% roztworu agarozy rozpuszczonej w wodzie. Sferoidy z komórek raka okrężnicy (HT29) przygotowano przy użyciu 96-dołkowych płytek z dnem w kształcie litery U i po 3 dniach wzrostu zebrano je, przemyto w PBS i dodano do mieszaniny żelowej. Pożywkę hodowlaną dodano na wierzch żeli zawierających sferoidy i pozostawiono je w temperaturze 37°C przez noc lub na kilka dni w celu sprawdzenia interakcji między sferoidami a agarozą. Rozmiar sferoid sprawdzono za pomocą mikroskopii optycznej i co ciekawe, sferoidy rosły w żelu agarozowym i modyfikowały otaczający obszar.
2% agarose gel was prepared from a stock of 4% agarose solution dissolved in water. Spheroids from colon cancer cells (HT29) were prepared using U-bottom 96-well plates, and after 3 days of growth, these were collected, washed in PBS, and added to the gel mixture. A culture medium was added on top of the gels containing spheroids, and they were left at 37°C overnight or for several days to verify any interaction between spheroids and agarose. The size of the spheroids was checked by using optical microscopy, and interestingly the spheroids kept growing in the agarose gel, and they modified the surrounding area.
